Yes! Black, white, and grays do count as design colors.

Printing on colored garments will cost more than printing on white or some grays. Please specify in comments if you want white or gray apparel.

Colors count in screen-printing because each one requires a unique screen. Lower your price by designing with one or two colors per side.
